Description
How well does your LC-MS workflow measure low-abundance glycans?
Detecting a glycan and measuring it reliably are different questions. The NeuGc Glycomics Benchmark Set helps you investigate both, using a defined series of complex N-glycan mixtures.
The set combines Neu5Gc-enriched N-glycans with a human serum glycan matrix. As the target glycan contribution changes across the series, you can assess how the measured response changes in a background relevant to plasma and serum glycomics.
What you can assess
- Sensitivity: investigate where Neu5Gc-bearing N-glycans become distinguishable from background
- Linearity: assess whether measured response changes proportionally across the series.
- Relative quantitation: identify the range over which individual target glycans can be measured consistently and proportionally.
- Workflow comparison: compare analytical approaches using the same benchmark material.
With suitable replicate measurements and acceptance criteria, the series can help establish detection and quantitation limits for individual glycans under your analytical conditions.
A practical starting point for glycomics
If your laboratory already has an LC-MS system and is introducing glycomics, the set provides released glycan material for assessing an analytical workflow. You can focus on separation, acquisition and data analysis from the point of injection, without first developing glycan release from a biological sample.
A benchmark for established laboratories
Use the set when comparing methods or systems, investigating low-abundance glycan measurements, or assessing the effect of changes to your workflow. The material also supports routine quality-control measurements and LC-MS installation performance checks.
Why a human serum glycan matrix?
Plasma and serum glycomics experiments involve complex glycan mixtures and a simple dilution curve of an single standard do not accurately represent the effects encountered when analysing these mixtures.
This set provides a sample context relevant to those experiments: Neu5Gc-enriched target glycans are combined with a human serum-derived glycan background. It complements purified structural standards by helping you assess relative response within a complex glycome.
Resuspend, inject and compare.
The sample preparation is already complete. Follow the product instructions to resuspend the samples for your analytical workflow, acquire the series, and compare the responses of your target glycans.
What’s included?
- Six tubes of dried, reduced N-glycan alditols.
- Each tube contains the glycan equivalent of a release from 500 micrograms of glycoprotein.
- Vial mixture ratios, a Skyline assay and reference data.
Dried samples are stable sealed at room temperature for six months. Record the resuspension and injection volumes, and begin with a low loading to establish an appropriate amount for your system.
